Posture is the window to the spine, so poor posture can negatively affect ones long-term health.
When one sits for prolonged periods without proper support it is easy to slouch. Rounding of the lower back and losing the normal curve in your lower back can cause a great deal of discomfort and spinal misalignment.
Here are some good sitting techniques that might be helpful:

A small cushion or a rolled up towel placed behind the lower back can help maintain a proper curve.
Make sure when you sit that your knees are higher than your hips.
When leaning forward to work at a desk, bend from the hips.
Keep your neck and back in a straight line.

When one stands for long periods of time, one wears out the muscles in the hips, lower back and abdomen, which tends to pull the pelvis forward straining the lower back muscles. Here are some good standing techniques:

Use a small footstool. Alternate resting one foot on the stool every few minutes. This keeps the blood flowing and the back's curve intact.
Always keep flexed
Transfer weight often.
Move around plenty - don't allow fixation in one position.
